Melanocortin receptors comprise five subtypes with distinct tissue distributions and functions. Melanotan-2 is described in the literature as a non-selective agonist that engages several of these subtypes, including MC1R, MC3R, MC4R, and MC5R. MC1R is the subtype most directly linked to melanin production in skin cells. Because the compound is not subtype-selective, its observed effects in experimental settings are generally attributed to activity across multiple receptor pathways rather than to a single target.
Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.

Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
== Early life and education == He was born in St. Paul, Minnesota on May 28, 1911. Nier showed an early ability in mathematics and science, coupled with an aptitude for craft and mechanical work. Nier's German immigrant parents had little education or financial resources but their determination for his development meant that he was able to attend the nearby University of Minnesota. Though he graduated in electrical engineering in 1931, the lack of engineering jobs during the Great Depression encouraged him to take up graduate study in physics.
==== Food Distribution and Food Insecurity ==== Organizations that are founded to target food insecurity have relied on food distribution to serve people in need of regular nutritious foods. Food banks are the proper term to call these kind of organizations where they use "government sector, private sector, and civil society" to distribute and recover food that will ultimately go to waste. In the United States, there are many organizations all over the country that have the same similar goal however, there is not much effective collaboration between all organizations because it is challenging to coordinate efficient communication within one another. There is one large organization that focuses on distributing food to people who need it in the United States, Feeding America has over 200 food banks and 60,000 food pantries. There are more organizations that focus on distributing food that work independently and are not as big as Feeding America that contribute to distributing food to help with the issue of food insecurity.
== Modern use == Commercially available hardtack is a significant source of food energy in a small, durable package. A store-bought 24-gram cracker can contain 100 calories (20 percent from fat) and 2 grams of protein.
== History == The Open Insulin Project was started in 2015 by Anthony Di Franco, himself a type 1 diabetic. He started the project in response to the unreasonably high prices of insulin in the US. The project has been housed in Counter Culture Labs, a community laboratory and makerspace in the Bay Area. Other collaborators include ReaGent, BioCurious and BioFoundry.

== Art == The continuity of the deep fasciae within the human body inspired the artistic expression seen in the Fascial Net Plastination Project, which is prominently displayed at the Body Worlds exhibition in Berlin.
== Urea cycle disorders == Urea cycle disorders are rare and affect about one in 35,000 people in the United States. Genetic defects in the enzymes involved in the cycle can occur, which usually manifest within a few days after birth. The recently born child will typically experience varying bouts of vomiting and periods of lethargy. Ultimately, the infant may go into a coma and develop brain damage. New-borns with UCD are at a much higher risk of complications or death due to untimely screening tests and misdiagnosed cases. The most common misdiagnosis is neonatal sepsis. Signs of UCD can be present within the first 2 to 3 days of life, but the present method to get confirmation by test results can take too long. This can potentially cause complications such as coma or death. Urea cycle disorders may also be diagnosed in adults, and symptoms may include delirium episodes, lethargy, and symptoms similar to that of a stroke. On top of these symptoms, if the urea cycle begins to malfunction in the liver, the patient may develop cirrhosis. This can also lead to sarcopenia (the loss of muscle mass). Mutations lead to deficiencies of the various enzymes and transporters involved in the urea cycle, and cause urea cycle disorders. If individuals with a defect in any of the six enzymes used in the cycle ingest amino acids beyond what is necessary for the minimum daily requirements, then the ammonia that is produced will not be able to be converted to urea. These individuals can experience hyperammonemia, or the build-up of a cycle intermediate.
Before the inexpensive production of gelatin and other competing products, isinglass was used in confectionery and desserts such as fruit jelly and blancmange. Isinglass finings are widely used as a processing aid in the British brewing industry to accelerate the fining, or clarification, of beer. It is used particularly in the production of cask-conditioned beers, although many cask ales are available which are not fined using isinglass. The finings flocculate the live yeast in the beer into a jelly-like mass, which settles to the bottom of the cask. Left undisturbed, beer will clear naturally; the use of isinglass finings accelerates the process. Isinglass is sometimes used with an auxiliary fining, which further accelerates the process of sedimentation. Non-cask beers that are destined for kegs, cans, or bottles are often pasteurised and filtered. The yeast in these beers tends to settle to the bottom of the storage tank naturally, so the sediment from these beers can often be filtered without using isinglass. However, some breweries still use isinglass finings for non-cask beers, especially when attempting to repair bad batches. Many vegetarians consider beers that are processed with these finings (such as most cask-conditioned ales in the UK) to be unsuitable for vegetarian diets (although acceptable for pescetarians). According to global data in 2018, along with low-calorie beer and gluten-free beer, beers that are acceptable for strict vegetarians are expected to grow in demand in the coming years.
Symptoms in people with Treacher Collins syndrome vary. Some individuals are so mildly affected that they remain undiagnosed, while others have moderate to severe facial involvement and life-threatening airway compromise. Most of the features of TCS are symmetrical and are already recognizable at birth. The most common symptom of Treacher Collins syndrome is underdevelopment of the lower jaw and underdevelopment of the zygomatic bone. This can be accompanied by the tongue being retracted. The small mandible can result in a poor occlusion of the teeth or in more severe cases, trouble breathing or swallowing. The respiratory system of a child with Treacher Collins syndrome is the primary concern at birth, with other issues only addressed once respiratory function has been stabilized. Underdevelopment of the zygomatic bone gives the cheeks a sunken appearance. The external ear is sometimes small, rotated, malformed, or absent entirely in people with TCS. Symmetric, bilateral narrowing or absence of the external ear canal, is also described. In most cases, the bones of the middle ear and the middle ear cavity are misshapen. Inner ear malformations are rarely described. As a result of these abnormalities, a majority of the individuals with TCS have conductive hearing loss. Most affected people also experience eye problems, including coloboma (notches) in the lower eyelids, partial or complete absence of eyelashes on the lower lid, downward angled eyelids, drooping of upper and lower eyelids, and narrowing of the tear ducts.

==== Functional linkers as catalytic sites ==== Functional linkers can be also utilized as catalytic sites. A 3D MOF {[Cd(4-btapa)2(NO3)2] • 6H2O • 2dmf} (H34-btapa= 1,3,5-benzene tricarboxylic acid tris [N-(4-pyridyl)amide], dmf = N,N-dimethylformamide) constructed by tridentate amide linkers and cadmium salt catalyzes the Knoevenagel condensation reaction. The pyridine groups on the ligand 4-BTAPA act as ligands binding to the octahedral cadmium centers, while the amide groups can provide the functionality for interaction with the incoming substrates. Specifically, the −NH moiety of the amide group can act as electron acceptor whereas the C=O group can act as electron donor to activate organic substrates for subsequent reactions. Ferey et al. reported a robust and porous MOF [Cr3(μ3-O)F(H2O)2(BDC)3] (BDC: benzene-1,4-dicarboxylate) where instead of directly using the unsaturated Cr(III) centers as catalytic sites, the authors grafted ethylenediamine (ed) onto the Cr(III) sites. The uncoordinated ends of ed can act as base catalytic sites. ed-grafted MOF was investigated for Knoevenagel condensation reactions. A significant increase in conversion was observed for ed-grafted MOF compared to untreated framework (98% vs. 36%). Another example of linker modification to generate catalytic site is iodo-functionalized well-known Al-based MOFs (MIL-53 and DUT-5) and Zr-based MOFs (UiO-66 and UiO-67) for the catalytic oxidation of diols.
=== Second Military Occupation === Estrada Palma's presidency ended with the Little August War and his resignation in 1906. This began the Provisional Government of Cuba, a second period of military occupation by the United States in Cuba lasting until 1909. President Theodore Roosevelt was a highly active Freemason, but his first Provisional Governor William Howard Taft would not be initiated until 1909, several years after Taft left the country. No evidence exists to suggest that Taft was involved in the Brotherhood at this time. His successor, Charles Edward Magoon, was never a Freemason. In the 1908 Cuban general election, both of the leading Presidential candidates, from opposing political parties, were Freemasons; José Miguel Gómez of the Liberal Party, and Rafael Montoro, of the Conservative Party. They were featured on the cover of the Grand Lodge's bi-weekly magazine, La Gran Logia. From the leadership of the new independent Cuba had emerged two oppositional political ideologies for its direction, and the Grand Lodge of Cuba was then fractured in the same manner. A large portion of the Freemasonic leadership in Cuba promoted that "Masonic virtue," should be the guiding light for all of Cuba, not just for Freemasons. Like a half-century earlier, debates about if Freemasons should be involved in politics raged. On March 22, 1908, Grand Master Fernando Figueredo Socarás published a message complaining that the political situation in Cuba was hindering the work of the Grand Lodge, and that it had "corrupted everything, including Freemasonry...
Renovo Group plc was a biopharmaceutical company founded in 1998 and headquartered in Manchester, United Kingdom. It worked in the discovery and development of drugs to reduce scarring, improve wound healing and enhance tissue regeneration. Renovo does not currently have any marketed products. It aimed "to be first to market with a scar prevention pharmaceutical drug in the US and Europe" in approximately 2014. Following the failure of its last clinical candidate, Juvista, all 100 of Renovo's staff were laid off in 2011. Although the company stopped all pharmaceutical development, it continued as a financial provider. In August 2014, Renovo Group plc was renamed Inspired Capital plc.
=== The California Sterilization Act of 1909 === This law passed in California in 1909, authorized medical staff like doctors and medical superintendents to perform sterilization procedures on both men and women deemed as feeble-minded, whose mental diseases, IQ, and intelligence could be passed down to future generations. A survey in mental deviations in prisons, public schools, and orphanages in California institutions reported worriness of feeble-mindedness and relation of intelligence to previous delinquency record. In their survey, they found that California had drawn a large proportion of immigrants of undesirable types and would therefore recommend them to sterilization processes. Statistical evidence demonstrates that there the number of sterilizations were disproportionate to racial and ethnic minorities, such as people of low class and female gender. Research also suggests that Latina women were targeted at significantly higher rates than non-Latinas. A 2018 study by Novak and colleagues analyzed over 17,000 forms recommending institutionalized patients for sterilization between 1920 and 1945. The researchers found that Latinas had a 59% greater risk of sterilization compared to non-Latinas. These findings of disproportionate sterilization represent how eugenic thinking reinforced racial stereotypes that portrayed Latinas as inferior and unfit to reproduce.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
No regulatory agency has authorised melanotan-2 as a medicine for any indication. It circulates mainly as a research chemical or through unregulated channels. As a result, identity, purity and content are not independently guaranteed.
